What Makes Daxxify Last Longer
Understanding why Daxxify lasts longer than other neuromodulators requires a brief look at how neuromodulators work and what has traditionally limited their duration. All botulinum toxin type A products work by blocking the release of acetylcholine at the neuromuscular junction, the connection between nerve and muscle, preventing the muscle from receiving the signal to contract. The toxin binds to the nerve terminal and cleaves SNAP-25, a protein essential for the vesicular release of acetylcholine. Once SNAP-25 is cleaved, the junction is blocked until the body regenerates new SNAP-25 protein and re-establishes the connection.
The duration of neuromodulator effects depends on how long it takes for this regeneration to occur, which varies between individuals and is influenced by factors including metabolic rate, muscle activity, and individual neurological characteristics. Standard neuromodulator products use the same core toxin with an albumin carrier protein that has a relatively short half-life in tissue.
Daxxify uses a novel peptide carrier protein instead of albumin. This peptide carrier, a synthetic peptide derived from research on the underlying mechanism of botulinum toxin binding, enhances the attachment of the toxin molecule to the presynaptic membrane and slows its clearance from the tissue. The result is prolonged occupancy of the neuromuscular junction and extended duration of effect.
The Clinical Trial Results
Daxxify’s FDA approval was supported by clinical trials involving approximately 2,700 participants. In the pivotal trials for glabellar line treatment, the median duration of response was approximately six months. At six months, a significant proportion of trial participants maintained a response, meaning their glabellar lines were still rated as none or mild on a validated scale. Some participants maintained their response for nine months.
These results represent a meaningful extension compared to the typical three to four-month duration of standard neuromodulators. For patients who have experienced consistently shorter durations from standard products, which some patients do, the extension may be even more significant.
Who Is a Good Candidate for Daxxify
Daxxify is appropriate for patients who are already maintaining neuromodulator results with standard products and want to reduce the frequency of treatment appointments. It is also a good option for patients who have noticed that their results from Botox or Dysport have been consistently shorter than three to four months, suggesting they metabolize standard products quickly.
Daxxify is currently FDA-approved for glabellar line treatment and is used off-label for other standard neuromodulator applications, including forehead lines, crow’s feet, brow lifting, lip flip, masseter reduction, and other areas. The extended duration is expected to apply across treatment areas, though the extent of this has been evaluated most thoroughly in the glabellar region.
Patients who are pregnant or breastfeeding, who have a history of neuromuscular disease, or who have certain other medical contraindications are not appropriate candidates for any neuromodulator, including Daxxify.

The treatment experience with Daxxify is essentially identical to treatment with Botox or Dysport.
